中文摘要 许多重要的农作物及经济动物都是多倍体物种,其中典型的例子是世界第三大粮食作物,同源四倍体栽培马铃薯。马铃薯营养丰富,与其他主要作物如水稻、小麦和玉米相比,对环境的适应性更强,丰产性更好。随着世界人口的快速增加、耕地面积的急剧减少、水资源的短缺以及膳食结构的改善,马铃薯在解决粮食安全、营养不良等全球焦点问题中扮演着越来越重要的角色。可靠的遗传连锁图是对基因组功能和进化进行分析的基础。在二倍体物种中构建遗传连锁图已成为常规研究手段;但在同源多倍体中,由于其减数分裂过程中染色体的分离和重组比二倍体复杂得多,此外越来越多的证据表明多倍化和其后多倍体基因组的结构与功能均经历一个高度动态的变化过程,使得其遗传连锁图构建的理论与方法学几乎是空白。本项目成功地建立了新的利用显性或共显性DNA分子标记构建同源四倍体物种遗传连锁图的理论模型与统计方法。该模型准确考虑了四体遗传中等位基因分离重组的重要特征,其可靠性与有效性通过对大规模计算机随机模拟数据和同源四倍体马铃薯异交群体AFLP和SSR分子标记数据的分析获得肯定。整合项目科研进展撰写的科学论文已被国际知名的综合性刊物PNAS接受发表。
英文摘要 Many evolutionarily, economically and agriculturally important plants and animals are autopolyploids. A typical example is cultivated potato, an autotetraploid species and the world’s third most important food crop. Because it provides a nutritious food supply and needs less demanding environment to grow but produces much higher yield than other major crops such as rice, wheat and maize, it is recently recognized as the vital food security crop when the world faces a fast increase in population but quick shrinkage in field area for crop cultivation. The availability of reliable genetic linkage maps is crucial for functional and evolutionary analyses of a genome. Construction of genetic linkage maps has long been a routine exercise in diploid species owing to the well established theory and method of genetic linkage analysis in these species. However, many evolutionarily, ecologically and/or agronomically important species are autopolyploids, with autotetraploidy being a typical example. These species undergo a much more complicated chromosomal segregation and recombination at meiosis than diploids. In addition, emerging researches have shown evidence of highly dynamic changes in the structure of the genome at the polyploid compared to the diploid level. These indicate the inappropriateness of the theory and methods of linkage analysis in diploids for use in tetraploid species and cast doubt on methods to approximate linkage maps of an autotetraploid by that of its diploid relative. Thus, there is a gap in the theory and methodology of map construction in these important species. The present research project successfully develops a novel theoretical model and statistical framework for multi-locus linkage analysis in autotetraploid species for use with dominant and/or co-dominant DNA molecular markers. The theory and methods take appropriate account of the essential features of allele segregation and recombination under tetrasomic inheritance and the major challenges in statistical modeling and analysis of the marker data. We validated the method and explored its statistical properties by intensive simulation study and demonstrated its utility by an analysis with AFLP and SSR marker data collected from an out-bred autotetraploid potato population. The scientific paper that reports the advances is accepted for publication in the world’s leading multidisciplinary journal Proceedings of National Academy of Sciences USA.
